How much of a difference in handling is there between a single seater and a sports prototype (as in LMP)?
I'd assume that they are similar due to the monococque construction but thats about all I know.

Probably depends...
Most single seaters are designed for short sprint races, whereas most sports prototypes are for longer (endurance?) racing.
Therefore it stands to reason that the single seater would be more on edge, as comfort, durability and relaxation aren't as important.
Also the LMP would almost certainly weight more, so it wouldn't be quite as direct or quick to react.
But as I haven't even sat in an LMP car I'm not the man to answer it definitely.
In terms of 'handling', you can adjust both of course.
But the driving difference is all down to weight.
Single seaters are the ultimate athletic instant responsive machines where you don't really 'think' about getting them along (unless they're low powered). When they are good, they are simple extensions of the driver's reactions.
LMP's are a more 'substancial' car with less grip, less atheticism in braking and accelerating as well as changing direction, using considerably more energy in getting along.
